The air pressure in isolation rooms depends on what you are trying to do. Negative pressure is used when you have something in a space that you don't want to get out and positive pressure is used when you have something outside of the space that you don't want to get in.
Requirements for how many isolation rooms are needed in a facility varies depending on your risk assessment.
